Total hemoglobin (HbA) and HbA1c have the same nonspecific absorption rate to latex particles. This is the basis of the first reaction in which sample is mixed with the R1 reagent (uncoated latex particles in buffer). The HbA1c that binds to the particles is proportional to the %HbA1c present in the sample.
Reagent R2 contains mouse antihuman HbA1c monoclonal antibody and goat antimouse IgG polyclonal antibody. Agglutination complexes will be formed from the interaction of the HbA1c bound to the latex particles with the respective antibodies. The agglutination complexes are measured as an increase in absorbance. The measured absorbance is proportional to the HbA1c bound to the latex which is, in turn, proportional to the %HbA1c present in the sample.
